Additional release information

SIRIUS 6.2 introduces a set of new features for the annotation of transformation products and the validating of structure database search hits using spectral libraries. This includes:

  1. integration of Biotransformer 3 for the generation of custom transformation product databases,
  2. a very fast spectral library analogue search, and
  3. the advancement of the "Substructure Annotation" view to seamlessly display mirror plots between the measured MS/MS spectrum and an analogue library spectrum. Fragment peaks in both spectra are annotated with substructures of the CSI:FingerID structure database hit, and the reference compound, respectively. This is a major difference to common analogue search where the structure of the measured compound remains unknown and fragment peaks have no structure annotation.

To provide these features, SIRIUS project and custom structure database schemas have been extended. This makes a conversion of existing projects and custom databases necessary to use their full potential with the new release. Please, read more about current compatibility below.

Major Changes and Known Compatibility Issues:

  • Compound Timeout: In SIRIUS 6.2, a compound timeout is set as default for molecular formula annotation. This timeout prevents computationally expensive features from getting "stuck" and massively slowing down the whole workflow. However, as a result, a few features may remain uncomputed. You can find "TimeoutException" warnings with corresponding feature IDs in the log. You can rerun computations later for such instances. Alternatively, you can set the "Compound timeout" (advanced settings) to 0 to disable it.
  • Existing Projects with Spectral Library Hits: For projects that have been created with previous SIRIUS versions, mirror plots in the "Substructure Annotations" and "Library Matches" views are not displayed. Metainformation of library matches is also not shown.
  • Existing Custom Databases with Reference Spectra: Custom databases that include reference spectra and have been created with previous SIRIUS versions are used in SIRIUS 6.2 only as structure databases, not for spectral library search. To use the full potential of your spectral libraries, please reimport the spectra with SIRIUS 6.2.

The ability to convert your projects and custom databases (that include spectral data) for full use with new SIRIUS versions will be provided in a coming release.
For SIRIUS 6.2, we switched to JCEF browser for rendering JavaScript-based visualizations. A Mac build will be provided after compatibility issues with the JCEF browser have been fixed.
